You will not need to equip or activate the stone in any way and just need to keep it with you in your inventory to have the Steeped in Bliss effect active.

The best way to get the Resonance Stone in Baldur’s Gate 3 is to search every nook and cranny of the Mind Flayer Colony during Act 2. There is no definite way of getting the Resonance Stone in the game but searching the Mind Flayer Colony should be your best bet.
